COLCHICINE


 








Definition: Colchicine is an alkaloid found in autumn crocus, a plant from the Liliaceae family (Colchicum autumnale).

Action: Colchicine blocks cell division by binding to the spindle fibers and causing their rupture, having as consequence accumulation of cells and metaphase. Colchicine acts by specific binding to microtubules, similar to what happen with N - deacethyl - N - Methylcolchicine (Colcemid - Ciba) to Vincristine and Vinblastine. For this reason, the change of medium may retrocede the process, allowing the culture to continue. As the molecular weight of colchicine is 359.43 and the concentration provided by VITROCELL is 4X 10-5M, it is used 0.05 to 0.1 ml for 5 to 10 ml of culture medium, for 30 minutes to 2 hours. This variation in time proceedings interferes in the concentration of the cromossomes, which increases as time elapses. Another factor that may be taken into account is that a longer time increases the number of metaphases, although causes minor distention of the cromossomes.

P.S.: Colchicine is a photosensitive substance, so avoid exposure to light as it may alter its chemical structure and result in loss of its properties.
